show Abstracthide AbstractThis project is to estimate the full-length amino acid sequences of Mesostigma viride (M. viride) photosynthetic proteins with high reliability using the full-length cDNA sequences obtained by an isoform sequencing (Iso-seq) analysis. First, the isoform sequencing (Iso-seq) analysis of M. viride mRNA was done with a PacBio sequel to obtain full-length cDNA sequences. The mRNA RNA was extracted from whole cells of M. viride (NIES-296), which was cultured under long day photoperiod (14h light / 10h of dark). The raw data was processed using SMRT Link v6.0 (Pacific Biosciences) and then further processed by IsoSeq3 tools (version 3.1). The full-length amino acid sequences of M. viride with a minimum length of 100 amino acids were estimated by a TransDecoder program from the high-quality consensus full-length cDNA sequences obtained by the Iso-seq analysis.
